The CCIB DNA Core, founded in 1995, is a well-established major research core laboratory within the Center for Computational and Integrative Biology of Massachusetts General Hospital Boston. The Facility provides a wide range of state-of-the-art services and specialized expertise in genomics, molecular biology, and laboratory automation to the greater Partners research community.
As the need for high-throughput research efforts is growing, the demand for laboratory automation solutions increases. To meet this demand, the Laboratory Automation unit has been designed as a flexible entity within the CCIB DNA Core to extend the benefits of laboratory automation to a diverse user group by providing standard and custom medium- to high-throughput sample processing services for various molecular biology and genomics applications. One of the many unique capabilities of the group is its proficiency in special automation process design and implementation.

- 96-well Plasmid DNA Preparation
The Laboratory Automation group has developed a purification system to isolate high-quality plasmid DNA in 96-well format. The purified plasmid DNA is suitable for a broad range of downstream applications, including transfection, in vitro expression, automated fluorescent DNA sequencing, and cloning. This plate-based DNA preparation service is offered as a stand-alone service or in combination with our DNA sequencing options.
- Bacterial Colony Picking & Archiving
Our well-trained technologists of the Laboratory Automation team will organize the clones of your library. Equipped with a Hudson RapidPick Complete Workstation, we offer automated colony picking and archiving, library re-arraying and library duplication services. Customized options are available.
- cDNA Library Screening
The Laboratory Automation group is now providing a high-throughput screening service utilizing a new plasmid-based, genome-wide human cDNA library. This new expression library was created in an extensive collaboration between Dr. Brian Seed's Laboratory and the CCIB DNA Core Team. The current library version consists of ~20,000 sequence-verified, full-length cDNA clones. The proprietary vector system (developed in the Seed Lab) allows easy shuttling of the individual cDNAs between different expression vectors via Cre/loxP-mediated recombination.
Please note that the CCIB DNA Core provides cDNA clones only within the context of a high-throughput screen and does not supply individual cDNAs (ORFs) or subsets of cDNAs (ORFs) for other purposes.

- Custom Automation Projects
The Laboratory Automation group provides customized automation support for a diverse range of research projects. A successful outcome for each individual custom project is achieved by a close collaboration between the research investigator and the dedicated scientists of the Automation team. The investigator is responsible for the experimental design as well as the development of a working manual version of the protocol, process or assay, which has to be semi- or fully automated. The Core’s experienced team will identify and develop tailored automated solutions to meet the project-specific medium- to high-throughput needs. Custom automation projects include screening, fluid handling, colony picking and sample preparation services.

- Mouse Genotyping
The Laboratory Automation group of the CCIB DNA Core provides a fast, accurate and reliable service for customized Mouse Genotyping. Since its launch in 2011, the PCR-based process has been continuously optimized and further streamlined by incorporating the use of robotic liquid handling workstations. The well-established service allows investigators to submit tail biopsies and primers and receive the genotyping results in form of agarose gel images in return. The use of laboratory automation enables us to process a large sample volume in a short period of time and to offer highly competitive pricing. Service is available for both individual samples and 96-well plates.

- siRNA Library Screening
The CCIB DNA Core houses the Silencer Select Human Genome siRNA Library V4 (Part Numbers 4397922, 4397924, and 4397923), which targets a genome-scale collection of 21,585 human genes. The Laboratory Automation group provides a high-throughput screening service utilizing a pooled version of the library. The pooled library consists of a total of 248 plates (96-well format), with three unique, non-overlapping siRNAs per well targeting the same gene.
