Core Summary:

The CCIB DNA Core, founded in 1995, is a well-established major research core laboratory within the Center for Computational and Integrative Biology of Massachusetts General Hospital Boston. The Facility provides a wide range of state-of-the-art services and specialized expertise in genomics, molecular biology, and laboratory automation to the greater Partners research community.

The CCIB DNA Core has the expertise to perform specialized services for investigators who are considering genotyping, DNA profiling, and mutation detection techniques for their research. Our experienced team - staff members from both the Sequencing group and the Laboratory Automation group - are providing fast, reliable, and cost-effective solutions for genetic analysis applications such as DNA Fragment Analysis (Microsatellite, MLPA) and Mouse Genotyping.

    The Core’s Sequencing group offers a fast and reliable automated ready-to-run DNA fragment analysis service where fluorescently labeled DNA fragments are electrophoretically separated using the ABI3730xl Genetic Analyzer for accurate sizing, consistency, and minimum run-to-run migration variations. Final data interpretation is available as an add-on service. Quality DNA sizing, allele calling, and relative quantitation is performed using the GeneMapper v4.0 analysis software package. Service is available for both individual samples and 96-well plates.

    - Microsatellite Analysis (short tandem repeats (STRs), variable nucleotide tandem repeats (VNTRs))
    - MLPA Analysis (multiplex ligation-dependent probe amplification)

  • Mouse Genotyping

    The Laboratory Automation group of the CCIB DNA Core provides a fast, accurate and reliable service for customized Mouse Genotyping. Since its launch in 2011, the PCR-based process has been continuously optimized and further streamlined by incorporating the use of robotic liquid handling workstations. The well-established service allows investigators to submit tail biopsies and primers and receive the genotyping results in form of agarose gel images in return. The use of laboratory automation enables us to process a large sample volume in a short period of time and to offer highly competitive pricing. Service is available for both individual samples and 96-well plates.

    Service Features include:
    - Extraction of high-quality DNA from tail clips
    - PCR-based Genotyping assays
    - On-site synthesis of custom PCR primers
    - Cost-free transfer of customer-validated assay(s) to the Core Facility
    - Weekly processing with fast turnaround
